Procedures of Streak Plate Innoculation Techniques are:

1. Sterilize the innoculating loop untill it is red hot and cool it.

2. Insert the loop into tube/ culture bottle and remove some innoculum.

3. Immediately streak the innoculating loop very gently over a quarter of plate using back and forth motion.

4. Flame the loop and cool again,extend the streaks into the second quarter of the plate.

5. Repeat step 4 to streak into the 3rd quarter and center fourth area of the plate.

Colony are calculated on the basis of:

1. All colonies should have the same general appearance.

2. If there is more than one type, each colony should be streaked again.

Procedures of gram staining:

1. Apply a primary stain ( crystal violet) to hot- fixed bacterial culture.

2. Addition of Iodide, binds to Cristal violet and holds it in the cell.

3. Rapid decolourisation by acetone or ethanol.

4. Counterstaining with safranine.

Bacteria differentiation techniques: Bio-, sero-and phagetyping, antibiotic susceptibility test, immunoblotting, polyacrylamide gel electrophoresis,etc.
