The transformation in mice observed by Griffith could also occur in vitro - Griffith injected live nonvirulent strain and heat killed virulent strain in mice and then he found that mice killed this means that transforming principle from virulent strain is transferred into nonvirulent strain and mice get pneumonia. Afterwords Avery, McLeod and McCarthy extracted transforming principle from virulent strain and added to nonvirulent strain invitro and by transformation that active principle is transferred into nonvirulent strain and then they injected into mice and mice was dead. This transformation can occur in vitro is concluded by Avery, McLeod and McCarthy.

C. DNase destroyed the transforming principle- To conform the transforming principle, Avery, McLeod and McCarthy treated transforming principle separately with RNase, Proteinase and DNase and they found that RNase and proteinase does not affect transforming principle and mice was dead . But transforming principle treated with DNase shows no death of mice confirms that DNase degraded transforming principle so that nonvirulent strain is not changed into virulent strain. This confirms that transforming principle is DNA.
