A 3000 bp circular DNA was treated with both HindIII and EcoRI restriction enzymes. EcoRI cuts at position 100. HindIII cuts at positions 500 and 2000. Which of the following represents the fragment that would be observed if the restricted DNA were analyzed using agarose gel electrophoresis?

Option A is the correct option.

We have a 3000bp circular DNA, when treated with HindIII and ECoRI restriction enzymes. ECoRI cuts at 100 position. HindIII cuts at positions 500 and 2000.

When ECoRI cuts at position 100 - we get that band of 100bp.

When HindIII cuts at position 500 - we get the band of 400 bp. Since we have a cutting site with ECoRI at 100 position.

When HindIII cuts at position 2000- we get the band of 1500 bp. Since we already have 100 bp band and 400 bp band. So, from 500 position to 2000 position we have a band of 1500bp.

Left out is the band of 1000bp. Band is from positions 2000 to 3000.

Total - 100+400+1500+1000=3000.

So, when we observe the restricted DNA using agarose gel electrophoresis we get 400, 100, 1000, 1500 bp bands.
