Solutions

Expert Solution

Solution 1

Given that,

Sotck solution of rabbit anti-HIV gp120 monoclonal antibody = 0.5 mg/ml (= 500ug/ml)

Required working concentration = 2 ug/ml in 1x PBS.

If we want to make 1ml of 2 ug/ml in 1x PBS,

Use the formula C1V1=C2V2

C1= Concentration of stock solution (0.5 mg/mL or 500μg/mL)

V1= Volume required (unknown)

C2 = Required concentration of solution (2 μg/ml)

V2= Required volume (1ml)

then, V1 = (C2 x V2)/ C1

= (2x1)/500 = 0.004ml or 4μL

ie take 4uL of 500ug/ml stock solution and dilute to 1mL using 1x PBS

To make 1ml of 2 ug/ml rabbit anti-HIV gp120 monoclonal antibody in 1x PBS, take 4μL of 500ug/ml stock solution and dilute to 1mL using 1x PBS

Solution 2

Given that,

Stock solution of rabbit anti-HIV gp120 monoclonal antibody conjugated to HRP = 0.5 mg/ml (500μg/mL)

Required concentration of 0.5 ug/ml

To make 1mL 0.5 ug/mL rabbit anti-HIV gp120 monoclonal antibody frim a stock solution of 0.5 mg/ml (500μg/mL),

Use the formula C1V1=C2V2

C1= Concentration of stock solution (0.5 mg/mL or 500μg/mL)

V1= Volume required (unknown)

C2 = Required concentration of solution (0.5μg/ml)

V2= Required volume (1ml)

then, V1 = (C2 x V2)/ C1

= (0.5X1)/500

= 0.001mL or 1μL

Dilute 1μL of stock solution into 1mL

To make 1mL 0.5 ug/mL rabbit anti-HIV gp120 monoclonal antibody frim a stock solution of 0.5 mg/ml (500μg/mL), dilute 1μL of stock solution into 1mL
