Design a study to investigate the sylvatic cycle (wildlife cycle) of a hypothetical mosquito-borne virus. It is known that small mammals such as rodents are the reservoirs. Use critical thinking skills as well as the CDC and the journal Emerging Infectious Diseases to determine how to design this study. Include the hows/whys/wheres of sampling including sample sizes. Include the hows/whys/and types of testing (e.g., PCR or antibody or other, etc.). Address any pitfalls or expected results of your study.

Answer:

  • Dengue is a viral disease transmitted by mosquitoes and caused by four viral serotypes belonging to the genus Flavivirus of the Flavivirus family.
  • In terms of illness and death, dengue is the most important viral disease transmitted to human by mosquitoes.

Mosquito Sampling:

  • Because many arbovirus of medical and veeterinary interest circular in the study area, used variety of sampling methods to collect a wide range of mosquito species.
  • Mosquitoes were then sampled by using human landing catches, CDC light trap with or without CO2, and animal bait traps.
  • Samples from these tree holes were stored in plastic boxes and flooded in the laboratory for egg hatching.
  • Virus isolation were performed on AP61 mosquito cell lines, as described by Digoutte et al. Virus was identified by using immunofluorescence with a specific fixation or neutralization tests.
  • The true infection rate estimated number of dengue virus positive mosquitoes per 100 mosquitoes tested
  • Nucleic acid detection: RNA is heat labile and therefore specimens for nucleic acid detection must be handled and stored according to the procedures described for virus isolation.
  • RT-PCR: In situ RT-PCR offers the ability to detect dengue RNA in paraffin- embedded tissues.
  • Extraction and purification of viral RNA from the specimen can be done by traditional liquid phase separation methods but has been gradually replaced by silica based commercial kits that are more reproducible and faster, especially since they can be automated using robotics systems.

Malaria is a mosquito-borne disease caused by parasites of the obligate intracellular Apicomplexa family, the most deadly of which, Plasmodium falciparum, prevails in Africa. Malaria imposes a huge health burden on the world’s most vulnerable populations, claiming the lives of nearly a million children and pregnant women each year in Africa alone. Although there is keen interest in eradicating malaria, we do not yet have the necessary tools to meet this challenge, including an effective malaria vaccine and adequate vector...

1) Is the following study design an anecdote, experiment, or observational study? You'd like to investigate if what color shirt fans wear to a game help the Le Moyne Women's Soccer team score goals? You attend every home game. You write down how many fans attended the game, what color shirt they wore, and how many goals the soccer team scored. 2) Among the 268 words in the Gettysburg Address, 99 contain at least 5 letters. Assume that the mosquito population across the southeast United States begins carrying the Zika virus. South Carolina legislators are concerned about the impact on pregnant women. A screening test (Test X) is required for all women of child-bearing age during annual appointments with their Ob/Gyn. The projected population to be screened in year 2020 is 100,000. State legislators want to know how many people will potentially need specialized medical care due to a positive test. You have the 2018 testing...
