Consider a colony of E. coli cells in which the gene for the CRP protein is deleted. Describe the effect of that this deletion will have on the arabinose operon in terms of levels of Ara A protein (one of the proteins encoded by a gene in the ara operon) when the cells are grown in media containing arabinose but no glucose. Explain your reasoning.

Answer:

  • The CRP gene is one that belongs to the pentaxin family and it is able to recognize many foreign pathogens along with damaged cell in the host.
  • It works in an attempt to eliminate the foreign and damaged cells from the humoral and effector system in the blood.
  • The gene is responsible for many host defense function that leads to many function like agglutination, capsular swelling, phagocytosis etc.
  • Arabinose operon is a very important regulation process and it is required for breakdown of the 5 carbon sugar L-Arabinose in E.Coli.
  • Ara A is one of the structural genes that encodes isomerase. In this case when the cell are grown in a media containing arabinose and no glucose they need to gain the energy for growth from arabinose by breaking it down.
  • However, if the gene for CRP is deleted, there will be no CRP and hence it will interfere with the function of the structural genes.
  • It was found that if there is a mutation in the CRP gene, it impacts all the structural genes but not AraA.
  • However it complete deletion is also going to slow down the process of Arabinose breakdown for gaining energy.

•In colony PCR of E. Coli, we were amplifying the GFP gene. In restriction digest, we were cutting out the GFP gene. The GFP gene is about 700 bp (base-pairs) and the entire plasmid including the GFP gene is about 3700 bp. Either draw out or explain where you would expect to see bands on the gel for each sample: •a) single digest, b) double digest, c) uncut miniprep, d) GFP PCR, e) control PCR

Which is likely to occur in E. coli cells that are grown in milk (Hint: Milk contains lactose sugar)? The lac operon is shut off, and the cells will not produce lactose-utilizing enzymes. The lac repressor is inactivated, and the cells will produce lactose-utilizing enzymes The lac operon is turned on, but the bacteria will not produce lactose-utilizing enzymes. ---- wrong
