could you have achieved a better separation on your TLC plate between aspartic acid and phenylalanine?
Yes. With mobile phase composition butanol–glacial acetic acid, in the ratio of 70:30. RF values could be 0.0 and 0.80 for aspartic acid (Asp - acidic amino acid - polar) and phenylalanine (Phe - Non-Polar) respectively. We know that polar substances tend to move slowly in TLC compare to non-polar substances. This rule is universal.
Please look at the page number 2 and Table Number 3 at below pdf published journal.

In another mobile phase n-butanol:acetic acid:water 3:1:1(by volume) mixture followed by ninhydrin stain the RF values are found to be Asp - 0.24 and Phe - 0.62 respectively. Please have a look at Reach Devices (Made in USA)
