If you wanted to measure expression of DNA at the level of specific transcribed mRNA sequences, what method could you use? (Describe quantitative reverse-transcription PCR (qRT-PCR).)

Solutions

Expert Solution

Reverse Transcription Polymerase Chain Reaction is a laboratory technique where the reverse transcription of RNA to DNA ( formation of cDNA) is happens and amplification of specific DNA target sites by using PCR. It is mainly used to measure the amount of specific RNA and it is done by the amplification reaction using fluorescence, and this technique is known as Real-time PCR or Quantitative PCR (qPCR). Combined RT-PCR and qPCR are commonly used for analysis of gene expression and quantification of viral RNA in research and clinical settings. RT-PCR has its own applications when it comes to laboratory and research. It can even detect very low copy number  of RNA molecules and it can be used in the diagnosis of genetic diseases and in the determination of the abundance of specific different RNA molecules within a cell or tissue as a measure of gene expression.

Genes are specific sequences of nucleotides that code for mRNA which may be translated into a protein. The nucleotide is the building block of nucleic acids, RNA and DNA. A nucleotide is composed of a pentose sugar, attached to one of five nitrogenous bases, guanine (G), cytosine (C), thymine (T), or adenine (A) and phosphate groups. The bases are covalently bonded through an ester bond between the hydoxyl group of one ribose and the phosphate group of another nucleotide. All...

I have a question about qPCR. How to measure an mRNA level of a gene by qPCR. If I perform a realtive quantification qPCR, first I need to convert mRNA into DNA template by Reverse transcriptase. then I perform a qPCR reaction. For example, comparing with the reference gene, the drug treatment lead to a decrease in the expression of gene of interest. However, I think that can only determine the gene replication is inhibited because the pcr reaction is...

Imagine that you have the DNA sequences from the intron of a gene in three species called A, B, and C. Species A and B are most closely related, while C is more distantly related. The sequences of A and B differ by 18 base pairs, A and C differ by 26 base pairs, and B and C differ by 28 base pairs. Fossils show that species A and B diverged about 1.2 Mya, but there is no fossil evidence...
