Answer :- Factor V ( Cofactor of Thromobokinase) is an anticoagulant produced in liver and converts Prothrombin (II - Inactive) to Thrombin (IIa - Active). Its molecular weight is 330 kilodaltons and plasma half life of 12 - 36 hours. Deficiency of factor V (Labile factor or Proaccelerin) can lead slowing of blood clotting process. The acquired deficiences of facor V are common than its congenital deficiences in newborn. The concentration of factor V in blood can be calculated by Factor V Assay, which is summarised as follows:-
1. Whole blood is collected in light blue top tube buffered with anticoagulant sodium citrate.
2. Mix blood with anticoagulant by gentle inversion.
3. Centrifugation of blood for 5 minutes in stat spin centrifuge.
4. Collect plasma from centrifuge tube and pour it into 4 ml BCS sample cup
5. Spin plasma for 5 minutes in same centrifuge.
6. Collect plasma after centrfuge and tranfer it into newsample cup for analysis.
