Discuss the pathological protein-protein interaction using the example of Sickle cell anaemia.
explain any structural changes/mutations that occurred to facilitate its pathological nature.

Solutions

Expert Solution

Protein-protein interactions are necessary to support proper functioning of the most basic molecular mechanisms. The normal PPI patterns get disturbed when an environmental stress or a disease state (such as sickle cell disease) disturbs the homeostatic state of an organism

The polymerization and depolymerization kinetics is altered by oxidative damage caused to the membrane and ionic channels of erythrocytes leading to formation of irreversible sickled cells

Nitric acid is responsible for maintaining homeostasis through regulation of smooth muscle tone, platelet activation and reductions in nitric oxide plasma levels

The superoxide anions (O2–).limits the availability of nitric oxide (NO) within the vascular wall. NO mediates vasodilation in a soluble guanylate cyclase when it diffuses through the cell membranes and bind to the nitrosylate proteins or heme-containing proteins. This is essential to several aspects of physiology and pathology.

Use the sickle cell anemia/malaria example to explain why alleles coding for negative traits can sometimes persist in a population over time. Your answer should include: what type of trait sickle cell anemia is (in terms of genetics) and which of the 2 alleles codes for sickle cell anemia? what the advantages and/or disadvantages of each genotype are for this trait? why the sickle cell allele is still present in many populations? Use paragraph format.
