Unlike SDS-PAGE which separates the denatured protein based on mass, size exclusion chromatography separates the protein molecules based on its mass and shape.Gel permeation chromatography can be performed using the gravity or attaching the column to a high-pressure pump. The large sample molecules will elute out first, and the smaller molecules will elute out latter. The size of the pores varies with different commercially available beads.
