Gelatinase test is used for testing the capability of certain bacteria to liquify the gelatin in the medium. The organism is inoculated on the agar and incubated at 250C. If the bacteria is positive, it will liquify the medium. To check whether liquification has occured, the medium after incubation is placed in ice bath or at a temperature of 40C.If the medium has liquified, at lower temperature it will get gelled again. This is a confirmation of liquification. So when placed in an ice bath, the gelatinase positive bacteria containing medium will form the gel again and will not stay in liquid form.
