To prepare DNA template with the concentration of 10 ng/μl:
Zach should add ____ μl of the original soil DNA (conc.: 250 ng/μl) and ______ μl of water into a tube to have final volume of 100 μl of the soil DNA that has the concentration of 10 ng/μl.
To prepare DNA template with the concentration of 10 ng/μl:
Zach should add 4 μl of the original soil DNA (conc.: 250 ng/μl= Stock) and 96 μl of water into a tube to have a final volume of 100 μl of the soil DNA that has the concentration of 10 ng/μl.
Concentration of DNA Stock =250 ng/μl
Required ethanol concentration (final concentration) =10 ng/μl of 100 μl
M1V1 (Concentrated) = M2V2 (Diluted)
250 ng/μl (Stock) * xul (?) = 10 ng/μl (Final concentration we need) * 100 μl (volume we need)
xul = 10 ng/μl *100 μl / 250 ng/μl
= 4 μl
According to dilution principle to get 100 μl of 10 ng/ μl of DNA we should have to take 4 μl of 250 ng/μl DNA Stock and add 96 μl of water (i.e. 4 μl of 250 ng/μl Stock and makeup it into 100 μl with water).
