Explanation - Most clinical and environmental specimens contain
several different microorganisms. To obtain a pure culture,
individual organisms must be isolated.
The most common method of isolation is the streak plate , in which
a sterile loop is inserted into a sample and streaked onto a plate
in a pattern, to obtain individual colonies.
Spreading - This technique typically is used to separate microorganisms contained within a small sample volume, which is spread over the surface of an agar plate, resulting in the formation of discrete colonies distributed evenly across the agar surface when the appropriate concentration of cells is plated.
In the case of pour plage - use of relatively hot agar carries the risk of killing some sensitive contaminants. Colonies of different species within the agar appear similar ā so it is difficult to detect contaminants.The reduced growth rate of obligate aerobes in the depth of the agar. Preparation for the pour plate method is time consuming compared with streak plate/and or spread plate technique
