You have purified genomic RNA from the following viruses: Zika, and murine leukemia viruses. You performed an experiment where susceptible, permissive cells were separately transfected with one pure molecule of viral RNA. Which RNA will lead to a productive viral infection? Discuss the reasons why one RNA succeeds and the other fails. Be clear and convincing
This study talks about transfection of cells with pure molecules of Zika and MLV . There is high probability of successful infection by Murine leukaemia virus than in comparison to Zika virus.
Now let us take both the cases one by one
Zika virus contains single stranded mRNA (+ve) . It belongs to the family Flaviviridae, One of the key factors about members of this family is they are mainly transmitted by blood sucking arthropods (Except Hepatitis C) and Aedes aegyptii in case of Zika. So, direct transfection may not succeed in case of zika virus. The need of an intermediate vector makes them less competent in transfection at the same time gives us another method to control them.
Murine leukaemia virus (MLV)- They are cancer causing RNA viruses of the family retroviridae. Here the replication happens by a DNA intermediate with the help of enzyme,reverse transcriptase. After getting converted to DNA they get integrated in host;s cell genome and are called Provirus. The emzyme reverse transcriptase is coded by the virus itself and the virus does not need any intermediate vector as well. So, there is hiigh probability of successful transfection by MLV.
