Microbes or bacteria are microscopic living forms. We can not see them by our naked eyes. We need to observe them only through microscopes of varying magnification power.

When we put bacterial microbes on the slide, we spread the culture as smear. Most bacteria being colorless the smear becomes transparent after drying on the slide.

Once colorless smear is dried on the slide. There comes 2 fold problem-

1st we can not locate easily on the slide that on which particular area of the slide the smear was spread (remember most bacteria are colorless)

2nd We can also overturn the slide because of abovesaid confusion.

Therefore, making the circle on the underside of the slide at the time of spreading the bacterial culture serves two purposes-

We can be sure that which area of the slide should be focused in the microscope for the bacterial observation

We can also be sure that which side of the slide contains bacterial smear and so, it avoids wrongful overturning of the slide.
