You receive an specimen in the lab from a patient with an infected throat. Chronicle the steps you would take to identify the bacteria present, starting with what stains, culture media, selective and differential. you would use and why?
Identification of bacteria from throat infected patient:
Firstly for isolating ,take the sample from patient by a throat swab.
Then place it on a slide then perform staining methods. Two types of staining are generally used they are gram staining and acid fast staining.
Gram staining is the most important and general method to determine gram positive and negative bacteria.
Where in gram staining if the bacteria in the microscope is in purple colour indicates gram positive and pink colour determines gram negative.To confirm this isolate bacteria in a culture medium,general media is nutrient agar.Then isolate the bacteria in selective media like mac conkey and eosin methylene blue agar which inhibits the growth of gram positive bacteria and supports the growth of gram negative bacteria.
(Acid fast staining is mainly used to isolate mycobacterium or other bacteria which is wax like cell wall .)
Then the bacteria which is positive in selective media is grown into differentiatial media like blood agar,citrate agar which is mainly used to differentiate by biochemical nature of bacteria.That is for example capability of lysis of blood and lactose fermented or non fermented.
By this help of tests we can identify the species of the bacteria.
Some.of the common bacteria found in throat is streptococcus and normal flora.
