What color do you expect a Gram-positive cell to be at each step of the Gram staining procedure? Why?
What color do you expect a Gram-negative cell to be at each step of the Gram staining procedure? Why?
Colour of gram positive bacteria | Colour of gram negative bacteria | |
1st step, addition of crystal violet stain | Crystal Violet is the Purple coloured stain and therefore the bacteria turns purple | same as Gram Positive Bacteria |
2nd step, addition of iodine | Crystal Violet iodine Complex are formed which are trapped in the mesh work of peptidoglycan layer and the bacteria appears purple | same as Gram Positive Bacteria |
3rd step, addition of alcohol | Crystal Violet iodine complexes are not washed off from the cell because peptidoglycan is thick and the bacteria is still purple | Crystal Violet iodine complexes are washed off and bacteria appears Colourless |
4th step, addition of safranine | Gram Positive Bacteria is still purple and will remain purple | Gram negative bacteria was colourless and now it will appear red |
