What color do you expect a Gram-negative cell to be at each step of the Gram staining procedure? Why?

Solutions

Expert Solution

Colour of gram positive bacteria Colour of gram negative bacteria
1st step, addition of crystal violet stain Crystal Violet is the Purple coloured stain and therefore the bacteria turns purple same as Gram Positive Bacteria
2nd step, addition of iodine Crystal Violet iodine Complex are formed which are trapped in the mesh work of peptidoglycan layer and the bacteria appears purple same as Gram Positive Bacteria
3rd step, addition of alcohol Crystal Violet iodine complexes are not washed off from the cell because peptidoglycan is thick and the bacteria is still purple Crystal Violet iodine complexes are washed off and bacteria appears Colourless
4th step, addition of safranine Gram Positive Bacteria is still purple and will remain purple Gram negative bacteria was colourless and now it will appear red
