Describe the two steps that are needed prior to expression of a protein of interest in Baculovirus instect system.

Solutions

Expert Solution

Baculovirus: this is virus known to infect many invertebrates. Immature(Larvae) of moth species are most common host and although they are capable to enter into mammalian cells and vertebrates but No ability of replication in vertebrates.

Its life cycle two distinct forms:--

1). ODV(Occulsion derived viruse) cause primary infection and occurs in protein matrix.

2). BV (Budded virus) cause secondary infection in insects.

Now come to the steps need prior to show infection or expression of protein in insect:---

1). Protein matrix To be dossolved in mid gut of insect:--- when a susceptible host insect attacks on such plant cantaminated with ODV and protein matrix of plant cell gets dissolved in alkaline environment of insect's mid gut. As a result ODV enters the insect cell. Viral transcription and replication occurs in cell nucleus.

2). Appearance of BV particles -- after transcription new BV particles are budded out from the basolateral sides to spread infection(expression of protein) symmetrically. During budding, BV aquires a loosely fitting host cell membrane with expressed and displayed viral glycoproteins.
