answer 1-7

1)What are the two main functions of lentivirus recombination?

2)What is the general purpose of the gag, pol, and env gene regions in a lentiviral vector?

3)Lentiviruses are useful because they lead to stable integration into the genome, can infect dividing and non dividing cells, have broad tissue tropism, and don't generate immunogenic proteins. What do each of these four phrases mean and why are they useful/essential?

4)What is the main difference between the first and second generation lentivrus systems?

Question 1- Lentiviral vectors have been done in gene therapy in order to cure desease like diabetes mellitus ,murine haemophiliaA,prostrate cancer choronic granulomatous desease and vascular disease.

It is usually created in a transient transfection system in which a cell line is transfected with three seprated plasmid.

Question 2- the three major proteins encoded within the retroviral genome gag,pol and Env.gag is polyprotein and is an acronym for group Antigen (ag) pol is the reverse transcriptase ENV in the envelope protein.

Question 3- signals and pathways ,uncoating process nuclear pore complexes are dynamic and respond to cellular stimuli this means that lentivirus can infect a graeter varity of cell types then retrovirus.

.lentivirus infection has advantage over than gene therapy methods including high -efficiency infection of dividing and non dividing cells ,long term stable expression of a transgene ,and low immunnogenicity lentivirus have also been successfully used for transfection of diabitic mice with the gene encoding PDGF .

Lentivirus are capable for infecting non dividing and actively dividing cell this means that lentivirus can infect a gretaer variety of cell types than retrovirus.

Question 4- first generation add genes packaging and envelope plasmids are generalized and appropriate for varried cell types and systems.

2nd generation lentiviral plasmids utilized the viral LTR promoter for gene expression.
