Why caspases 2 is crucial for the intrinsic pathway of apoptosis?

Solutions

Expert Solution

Caspase 2 play an important role in execution of apoptosis.

Different type of cellular stress ( DNA damage, microtubules disruption, oxidative stress) lads to activation of caspase-2 . Later this caspase two ( this is a cysteine protease) act on pro apoptotic protein BiD and cause release of its cleaved products iBid . This iBID interact with BAX and BAX present in mitochondria membrane ultimately cause pore formation and MoMP ( mitochondria outer membrane permeablization). Thus lead to burst of mitochondria and release of cytochrome c which later formation of  Apaf 1 and cause death. Thus caspase 2 is important in execution of death by MOMP and release cytochrome C.

The process has been explained in easier way in attached image below
