  • Simple stain- This type of stain is used to determine the shape, size, and arrangement of the microorganisms. It uses only one stain as the name states simple stain. This type of staining is used to show the presence of bacteria as opposed to the background.
  • Differential stain- This type of staining is done to differentiate between two different microorganisms. It uses more than 1 stain. This is done in detecting abnormalities in proportion of different WBC.
  • Negative stain- In negative staining the background is stained, leaving the actual specimen untouched. This method of staining is used for study and identification of aqueous lipid aggregates like lamellar liposomes, inverted spherical micelles,etc
  • Acid-fast stain- The acid-fast stain is a differential stain used to identify acid-fast organisms such as members of the genus Mycobacterium. They appear red in colour.
  • Flagella stain- The flagella stain allows observation of bacterial flagella under the light microscope. Bacterial flagella are normally too thin to be seen. The flagella stain helps in coating the flagella and making it thick enough to be seen under microscope.
