Strength of elecron microscopy

• Magnification and higher resolution - the resolution of electron microscopy images ranges upto 0.2nm, which is 1000xmore detailed than light microscopy
• diverse application- it canbe used in different fields like technology ,industry, biomedical science,and chemical science.
• high quality images- it can produce highly quality , detailed images, and analyse complex and delicate structures.

weakness of electron microscopy

• Inability to analyse live specimens- so biological specimens cannot studied through electron microscope, as electrons are easily scattered by other molecules in the air,samples must be analysed in the vaccum.
• Black and white image- only black and white image can be produced by electron microscope
• cost - electron microscope are expensive equipments. so it cannot applicable to low cost projects
• Artefacts- requires specialized knowledge of sample preparation to avoid
• size- it is larger in size requires more spaces in laboratory.
• training- specialized operators requires to handle the instrument

techniques of electron microscopy

   An electron microscope that uses a beam of accelerated electrons as a source of illumination.the wavelength of electrons can be upto 100000times shorter than that of visible light photons.TEM is a microscopy techniques in which a beam of light is transmitted through a specimen to form an image.uses high voltage electron beam to illuminate the specimen and create an image . the image is magnified and focused on to an image device ,such as flourescent screen , a layrer of photographic film or a sensor The electron beam is produced by an electron gun ,commonly fitted with tungsten filament cathode as an electron sources.

SEM produces images of a sample by scanning the surface with focussed beam of electrons. The electron interacts with atoms in the sample , producing various signals that contain information about surface topography and composition of the sample . the electron beam is scanned in a raster scan pattern , and the position of the beam is combined with the intensity of the detected signals to produce an image.
