A. Describe three important differences in the transcription of mRNA's between bacterial and eukaryotic cells.
B. In the synthesis of cDNA libraries for eukaryotic genes, reverse transcriptase can be used with a primer containing poly(T). Why is this NOT useful for prokaryotic systems?
A. Difference between transcription of mRNA's between bacterial and eukaryotic cells.
Eukaryotic transcription | Prokaryotic Transcription |
Have three RNA polymerase I, II, amd III | Have one RNA polymerase |
the RNA is first transcribed in the nucleus and then translated in the cytoplasm. | transcription and translation occurs simultaneously in cytoplasm |
Requires proteins known as transcription factors for the initiation. | Doesn’t require any proteins or other factors for the initiation of transcription |
B. In prokaryotes very less RNAs are poly adenylated as it leads to degradation and as the transcrition and translation occurs simultaneously in cytosol. In eukaryotes all coding mRNAs will have poly A tail for protection during tranfer from nucleus to cytosol. As there is no poly A tail in all prokaryotic mRNA we acnt use Poly T primer for reverse transcrition in prokaryotic system.
