Why is it important to quickly cool tubes filled with agar in an ice bath, when the inoculation of the brain heart infusion agar deeps?

Solutions

Expert Solution

Because to make oxygen gradient throughout the tube it is important to quickly cool tubes after mixing bacterial inoculum

Once the agar solidifies,an oxygen gradient is created with atmospheric oxygen concentration at the top of tube and little or no oxygen at bottom of the tube

The bacteria only grow in the part of tube where their oxygen requirements are fulfil and localised in that area

And by that we can find the oxygen requirements of that organism(which was our aim).

Using an ice bath, cool 10mL of saturated sodium bisulfite solution; then add, drop by drop, 5mL of 2-pentanone. Stir the solution until there is precipitation of a solid. Add 25 mL of ethanol to encourage precipitation; then filter the crystals and wash them with 5 ml of ethanol. The product is a bisulfite addition adduct. What makes the Bisulfite product soluble in 10% NaOH but insoluble in 10% HCl. What does ethanol do to the product in this reaction?...
