What is the main advantages of Capillary electrophoresis as compared to HPLC? What determines the elution order in capillary electrophoresis? How elution in CE differs from HPLC?

Solutions

Expert Solution

Main advantages of Capillary electrophoresis as compared to HPLC :

Capillary electrophoresis gives good resolution and helps to reduce band-broadening seen in the peaks as compared to high performance liquid chromatography (HPLC).

Other advantages of Capillary electrophoresis as compared to HPLC :

HPLC is costly and require large quantities of expensive organics. Capillary electrophoresis is cheaper and quicker.

Determination of the elution order in capillary electrophoresis :

Capillary electrophoresis separates ions depends upon dielectric constant but polarity is a "fair" measure of this ability

The more polar sample will retained on the stationary phase for longer time. So least polar compound will elute from the column first and then followed by order of increasing polarity.

Elution order of HPLC is governed by polarity.
In case of HPLC, least polar solute will elute first and the most polar solute will eluted at the last.
