  1. Below are 3 types of DNA crosslinking damage that can occur. Based on your understanding of DNA repair, what mechanism would you predict would repair each and WHY?
    1. Inter-Strand Crosslink:

  1. Intra-Strand Crosslink:
  1. Pyrimidine Dimer:

Solutions

Expert Solution

A. inter-strand crosslink damage is repaired by homologous recombination as the damage is in both strands and for this kind of repair uses template strand of another DNA is used for repairing.

B. Intra-strand crosslink is repaired by Base excision and nucleotide excision repair system. If only a single base is damaged BAse excision repair mechanism is used, But if 2-4 nucleotides are damaged Nucleotide excision repair system is preferred.

C. Pyrimidine dimer can be repaired by photoreactivation. Photoreactivation is a light-induced (300–600 nm) enzymatic cleavage of a thymine dimer to yield two thymine monomers. It is accomplished by photolyase, an enzyme that acts on dimers contained in single- and double-stranded DNA.

DNA mutations can occur in a variety of ways. Choose one of the ways that DNA mutations can occur that you are learning about this week, identify and briefly describe it. Then, choose a genetic mutation (it can be from your textbook, one you are aware of, or one you learn about through your research of this topic), and describe the type of mutation that it involves. Your cells contain machinery to detect and repair DNA damage to protect the fidelity of your genetic code. One form of damage detected by this machinery is cytosine deamination. Explain why replacing the C4 amine in cytosine with a hydroxyl could result in an altered DNA sequence upon replication. Hint: consider tautomerism. Please explain in as simple terms as possible.
