3- Surface treatment regimes (need to consider a method that allows for multiple zonally arranged functional groups within a continuous scaffold for the production of a hierarchical structure. Things to consider: chemical grafting, control of topography, attachment of biomolecules)

4- Preliminary In vitro test (define a series of in vitro tests that you would use to assess the suitability of your scaffold (or modified surface) for its given clinical target. You should include cell source and type, type of medium and describe a series of tests that would not only identify the cytotoxicity of the proposed material, but also be used to evaluate the phenotype of the adhered cell population. Time points and appropriate controls for specified methodologies )

1)It is help for bone repairing and transplanting in severe bone defects or disease.recent days,well developed methods in tissue engineering is to help for complete bone transplanting.studies on scaffold used to repair bones and bone regeneration.it is also used in engineering of cartilage.it can be used in the treatment of periodontal and alveolar defects.

it has good osteogenesis,biocompatibility,and security.applying scaffolds on clinical field show a better ability to facilitate to repair bone .

for the construction of of 3D saccffolds mainly use chilosten as it is considered as an excellent  polymer.other than this silicon rubber,polypropylene, and polymethylmethacrylate are the polymers used to make saccffolds. .

In vitro test will help to understand saccfold immunogenecity.and its primary tenocyes is assed using live dead staining.mean while,phenotypic retention is checked through real time PCR analysis of tenocytic genes.in in vitro  test mainly extra cellular matrix and skill like material are used.
