For the diagnosis of type 2 diabetes mellitus generally following tests are done:-
HbA1C,
Fasting blood sugar, and
Oral glucose tolerance test.
HbA1C :- To know the status of the blood glucose for the past two to three months which is very necessary in case of type 2 diabetes mellitus.
Fasting blood sugar:- To know the level of glucose in the blood after the fasting of 7- 8 hours and if the value of blood glucose comes 126mg/dl or higher on two occasions that means client is having type 2 diabetes mellitus.
Oral glucose tolerance test:- To know the blood glucose level after giving the oral glucose and if post 2 hours of the oral glucose given blood glucose level is 200mg/dl or more that means client is having type 2 diabetes mellitus.

Random blood sugar is primarily used for the diagnosis of the type 1 diabetes mellitus.
Hemoglobin level is used for the diagnosis of anemia not diabetes.
