It can be observed that there will be no much difference of reaction with normally 1:2000 dilution of crystal violet (intravital staining dye) on cell wall wall of bacterial organism even in gram positive or gram negative bacteria as substituting with Gram’s crytal violet has nealy similar reaction with bacterial organism cell wall especially with gram negative bacteria which retain purple colour with gram stain.

Gram negative bacteria are characterized by their inability to retain crystal violet stain, which is used to differentiate gram positive and gram negative bacteria. The three layers of the gram negative envelope include,

  • The inner cytoplasmic membrane
  • Middle peptidoglycan layer
  • Outer lipopolysaccharide layer

The middle peptidoglycan layer is sandwitched between the inner layer and the outer layer, which is very thin to retain the crystal violet stain.
