A) Given 37ul of a 100 mg/ml stock solution of Bovine Serum Albumin (BSA), diagram the most accurate procedure so you end up with at least 500ul each of 5ug/ul, 0.5 ug/ul, and 0.05 ug/ul working solutions of BSA.
B) Using the BSA working solutions created above, how much and which concentration would you pipet into three different centrifuge tubes so you end up with 20 ug, 2 ug, and 0.2 ug of BSA? (Note: remember to stay within the accurate range of your pipetman.)
Answer A)
For part 3,
Pippette out 50 uL of solution in part 2 and add 450 uL of solvent to make 500 uL solution.
Answer B)
For 20 ug solution, solution made in part 1 must be used.
Solution made in part 1 is 5 ug/uL.
So, 1 uL of above solution he 5 ug of BSA.
For 20 ug, solution to be added is (20/5) = 4 uL.
For 2 ug , solution made in part 2 should be used
Solution made in part 2 is 0.5 ug/uL.
So, 1 uL of above solution the 0.5 ug of BSA.
For 20 ug, solution to be added is (2/0.5) = 4 uL.
For 0.2 ug solution, solution made in part 3 must be taken.
Solution made in part 3 is 0.05 ug/uL.
So, 1 uL of above solution he 0.05 ug of BSA.
For 20 ug, solution to be added is (0.2/0.05) = 4 uL.
