i.e. Human platelet aggregometery assay :

Step 1 : Take blood sample of the subject

Step 2 : centrifugation of blood samples

Step 3 : take PPP(platelet poor plasma) in one cuvette, equates 100% light transmission

Take PRP(protein rich plasma) in another cuvette, equates 0% light transmission

  • For the calibration of aggregometer:

Step 4 : PRP is added to the aggregometry cuvette and warmed at 37°C until a steady baseline is achieved.

Step 5 : agonist added to cuvette & record the response

Step 6 : repeat the procedure with other agonists

  • Use of aggregating agents:
  1. Used to facilitate the aggregation of platelets
  2. Used to take the record of different respomses for the diagnosis of diseases
  3. Use to produce magnified response of platelet aggregation which is easily recordable and noticeable
  4. Eg. ADP, Epinephrine, serotonin, archidonic acid etc.
  • Use in disease diagnosis :

Platelet aggregation assay show different respomses with different agents by which diseases can be diagnosed

For e.g. Storage pool disorder shows primary aggregation with ADP, adrenalime amd collagen while defect in cox pathway shows absence of aggregation with archidonic acid.
