Solutions

Expert Solution

1. The tumor-suppressor genes are the genes that keep the cell growth in check. They stop the tumor causing genes or in other words the cell cycle initiating genes in check. By doing so they stop the uncontrolled cell division thereby suppressing the genes responsible for causing of tumor. p53 is a tumor suppressor gene and suppresses various cell cycle pathways when in normal condition. But it p53 was mutated then the cell cycle pathway will get activated and uncontrolled cell divisions will take place producing a tumor. Hence the correct answer from the given options is: A. When not mutated, most act as restrictors of cell proliferation.

2. C.Immunotherapy is an approach that tries to get the immune system more involved in the fight against cancer. In immunotherapy, the body's immune system response is heightened to counter attack the cancer causing agents and eliminate them. The other options mentioned such as gene therapy works by editing the cancer causing gene in-vitro and then reintroducing this gene into the host cell. The inhibition of cancer promoting proteins is done by the chemotherapy or radiation and this doesnt involve much of the bodys immune system reaction. The inhibition of angiogenesis is the inhibiting the supply of blood to the cancer cells thereby decreasing the amount of nutrient flow to cancer cells and hence starving the cancer cells. This also doesnt activate the immune system.

Retinoblastoma (Rb) is the model genetic system for tumor suppressor genes. Analysis of DNA from retinoblastoma patients revealed that Rb is a recessive autosomal condition with LOH; however, analysis of pedigrees segregating for the familial form of Rb indicates that the condition is inherited as an autosomal dominant trait. Explain the apparent inconsistency and define LOH.
