Compare and contrast SYBR Green versus TaqMan?
SYBR green:
• It is an asymmetrical cyanine dye used as nucleic acid strain in molecular biology.
• It binds to Dna forms the complex that is best absorbs at 497nm at blue light and produces(emits) green light.
• It preferably binds to double strand dna but can also strain Rna with lower performance than ssDNA(single strand).
• Used in several fields of biochemistry and molecular biochemistry.
• Used as dye for dna quantification in quantitative PCR (polymerized chain reaction).
• Used in labelling pure nucleic acid strain in dna in flow cytometry ad fluorescence microscopy.
Taqman:
• Its a hydrolysis probe to increase the specificity of quantitative PCR.
• It relies on the principle of 5' - 3' exonuclease activity of taq-polymerase to cleave a duel labelled probe during hybridization to target sequence and fluorescence based detection.
• Intrestingly the name originated of taqman is from video game that is taq-polymerase + pacman= taqman.
• The fluorescence detected in the quantitative PCR thermal cycler is directly proportional to the fluorophore released and the amount of DNA template present in the PCR.
• Its widely used in quantitative PCR in research and medical research like
• Pharmacogenomics, gene expression assays, HLA- genotyping,
• Dna quantification assays, bacterial identification assays, verification of microbiology results etc.
