What chemical you use to induce cell competent for transformation? What are three important features a vector should have?  Why it is important to have the recovery process during transformation? What is the next step after recovery process?

Solutions

Expert Solution

1. Calcium chloride is used to make bacterial cells competent for transformation.

2. A vector should have the following three important properties:

(a) Ori (Origin of replication) for initiation of replication of the plasmid

(b) Antibiotic resistance genes/selectable marker for selection of transformants and recombinants

(c) MCS (Multiple Cloning Site) / Polylinker site containing the restriction enzyme sites for insertion of the insert DNA/gene of interest

3. Recovery is a very important step as temperature applied during heat shock causes the cells to become stressful and thus requires recovery time.

During recovery, the transformed cells also start to express the transformed genes such as the antibiotic resistant genes.

4. The next step after recovery is to plate the recovered cells on a suitable antibiotic containing plate and incubate the same at 37 degree C to select for the transformants/recombinants.

Important: you are required to use only the functional features of Scheme; functions with an exclama- tion point in their names (e.g., set!) and input/output mechanisms other than load and the regular read-eval-print loop are not allowed. Nomenclature in Medicine Chemical nomenclature is an important skill that you will use outside of this class. In many ways, it is a primary skill needed in pharmacology and medicine. For example, a common imaging agent used for x-rays of the gastro-intestinal tract is barium sulfate. Despite the high toxicity of barium, barium sulfate (BaSO4) can be safely ingested because it does not dissolve well in water and passes through the intestines with little effect on the patient. In contrast,...

You have previously used radiation therapy to induce cell apoptosis in a mouse tumor model in which FGF is over-expressed. Recently, you notice that this therapy is no longer effective and the tumor continued to grow rapidly. Genomic sequencing revealed that those tumor cells have acquired a new loss-of-function mutation in the tumor-suppressor gene Pten, whose protein product normally acts as a phosphatase to dephosphorylate PIP3 into PIP2. Based on this information, you have an idea of why the tumor...
