A new gene called “Kmk” is suspected to play important role(s) in heart development of mouse.
What can be the important experimental observation to suggest Kmk is important for heart development in mouse and then design two experimental approaches with research plans that can help to prove that gene Kmk is really important for heart development in mouse.
If we observe in experiments that the mutation in the Kmk gene causes no development of heart then we can say that Kmk gene is necessary for heart development.
Two experimental approaches:
1. We will take two identical mouse embryo, In one of them the Kmk gene will be knocked out . If the Kmk gene is responsible for heart development, the Kmk knocked out mouse will not develop heart whereas the non knocked out mouse will develop heart. By this approach we can prove Kmk gene is necessary for mouse heart development.
2. The second approach may be that we will design a miRNA which will specifically bind to Kmk transcribed mRNA and therefore inhibit the expression of Kmk gene. We will take two identical mouse embryo, and in one of them we insert this miRNA. If the miRNA injected mouse do not develop heart while the non injected mice develop normal heart,then we can conclude that Kmk gene product is essential for normal heart development in mouse.
