Chapter T21, Problem 2P
In reversed-phase, the solutes will have a faster rate of migration when there is a less polar solvent present. The advantage of using the reversed-phase over the normal-phase chromatography is that the rate of exchange of the solute molecules between the two phases happens very quickly, allowing the substances to flow through the column much faster.
In normal-phase chromatography, the gradient elution will be from no polarity to gradually increasing polarity; in reversed-phase chromatography, the gradient elution will be from high polarity to gradually decreasing polarity.
