  1. Isocratic elution is where the composition of the mobile phase is unchanged during the entire elution process. Gradient elution is where the solvent strength of the mobile phase in increased during the elution starting with a solvent of relatively low solvent strength. Method development is easier with gradient elution chromatography than isocratic elution chromatography. Because, gradient elution decreases the retention of the later-eluting components so that they elute faster, giving narrower (and taller) peaks for most components.
  2. In recent years, the use of elevated temperatures in liquid chromatography has become more familiar, although many people still fear to increase the temperature because of unwanted side effects. In most laboratories, separations are usually performed at room temperature. In this setting, elevated temperature liquid chromatography would mean the temperature ranges from 40oC-60oC, whereas high temperature liquid chromatography extends from 60oC to 200oC.
  3. Dry lab is an educational exercise in which most of the hand on components a particular lab procedure are performed with the exception of those requiring use of a sink and biohazardous disposal.
