Successful DNA synthesis during the process of replication requires enzyme/s such as

A) Helicase
B) DNA primase
C) DNA ligase
D) DNA polymerase
E) All of the above

Solutions

Expert Solution

Ans.Successful DNA synthesis during the process of replication requires enzymes such as:-(E).All of the above,i.e.DNA helicase,DNA primase,DNA ligase & DNA polymerase.Function of these enzymes in DNA replication:-(1).DNA helicase :-DNA helicase also known as helix destabilizing enzyme.Helicase separates the two strands of DNA at the replication fork behind the topoisomerase.(2).DNA polymerase:-DNA polymerase are family of enzymes that carry out all forrns of replication. The main enzyme is referred to as DNA dependent DNA polymerase, since it uses a DNA template to catalyse the polymerisation of deoxynucleotides. The enzyme responsible for catalyzing the addition of nucleotide substrate to DNA in 5' to 3' directiion during DNA replication. The enzyme also performs proof- reading & error correction. There exist many different types of DNA polymerase, each of which perform different functions in different type of cells.(3).DNA ligase:-Re-anneals the semi-conservative strands & joins Okazaki fragments of lagging strands.(4).Primase:-Provides a starting point of (RNA) or DNA for DNA polymerase to begin synthesis of new DNA stand. These all enzymes are necessary for successful DNA synthesis during the process of replication.

DNA replication requires many enzymes to accomplish the process successfully. This results in a replication process that is known to be bi-directional, semi-conservative and semi-discontinuous. You discover a new prokaryotic organism that lacks the gene coding for an DNA polymerase I enzyme, and therefore this enzyme is not made in this species. Additionally, the DNA polymerase III enzyme from this organism lacks any 3’ to 5’ nuclease activity but has 5’ to 3’ nuclease activity. Based on what you know...
