Cultivation independent method

It is molecular method for microbial studies for its identity and presence within their environments.
This method is mostly used now a days due to its practical benefits, like its minimum time consumption and ability to identify great proportion in bacterial diversity which is difficult to identify in the cultivation dependent method (culture dependent).
This method mainly involves the DNA sequencing ( metagenome analysis of 16s rRNA ) collaborating with bioinformatics tool, FISH etc are more frequently used .

Culture dependent method

It is method of growing prokaryotes in a culture in laboratory set up.
In this method the microbes are let to reproduce in a designed culture medium and depending on the site the culture medium varies, that is ; if the infectious microbes in uterus is to be studied,then its tissue lining is taken and selectively cultured.
This method is comparatively less used now a days because of more time consumption and resource requirement, risk of contamination etc.
This method involves plate count, fatty acid methyl ester (FAME) analysis, impression techniques
