CRISPR Lab

Describe at least two other experiments that could be done to verify that chromosomal gene editing occurred in the bacteria.

Solutions

Expert Solution

Ans:- The two ways by which we can detect that gene editing have occurred or not.

1. western blot

The edited gene will not we able to synthesize the protein. So, if we do western against the protein that have been edited, there will be no band that will appear in the western blot. This confers that gene have been edited.

2. PCR followed by sequencing

PCR amplify the region using the flanking end primers that is edited. Means primers will be used in such a way one primer is upstream to site of editing and other to the downstream. Now, the amplified PCR product is send for sequencing, the sequencing results will say that whether your gene have been edited or not.

You can also use restriction enzyme, if you know the place where the gene have been edited. If the gene is edited then nuclotides sequence will be changed. Chosen such a enzymes that cut only your wild type DNA but not edited gene. Then compare the results that will confirm that whether your gene is edited or not.

Dear student if you find the answer convincing please do give the feedback.

Consider the experiments. Experiment 1: A study is done to determine which of two fuel mixtures allows a rocket to travel farther over a period of time. Rocket A, which requires additional equipment to keep it stable, is used to test one fuel mixture, and rocket B is used to test the other. Both rockets are identical aside from their mass. The results indicate that rocket B traveled farther than rocket A over the same period of time. Experiment 2:...
