Jacob and Monod proposed the operon model by analyzing mutations in the different components of the lac operon. Below are the mutations.

Repressor works in a trans manner, it means that repressor produced in plasmid can bind to the lac operon of the genome.
The operator works in cis-manner, it means that if there is a mutation in one operator of genomic operon then it will affect the expression of plasmid operon.
If the merodiploid condition has IS and OC then OC will show its effect
If LacZ is mutated then b-Galactosidase will not show its activity irrespective of lactose presence of absence.
If LacY is mutated then Permease will not show its activity irrespective of lactose presence of absence.
