What are the components/ steps of the genetic mapping in plants?
1. Fragmentation of genome: Fragmentation of the genome into overlapping clones is usually done by using rare cutting enzymes so as to produce large DNA fragments.
2. Preparation of a large fragment library: The fragments thus generated are ligated to cloning vectors of high capacity like YAC.
3. Overlapping clone identification: Two strategies are used to identify overlapping clones: one is clone fingerprinting and the other is chromosome walking. In the clone fingerprinting method, overlapping clones are identified based on common patterns of restriction enzyme fragments or repeat content observed between clones. In chromosome walking, the overlapping clones are identified based on hybridization of two overlapping clones to a particular STS marker.
4. Preparation of STS content map: Arranging the overlapping fragments based on the presence or absence of a signal for a particular probe.
