In the clone fingerprinting method, overlapping clones are identified based on common patterns of restriction enzyme fragments or repeat content observed between clones. In chromosome walking, the overlapping clones are identified based on hybridization of two overlapping clones to a particular STS marker.

4. Preparation of STS content map: Arranging the overlapping fragments based on the presence or absence of a signal for a particular probe.
