You are going to use column chromatography to separate a mixture of four dyes: 4-phenylazophenol, Scarlet-R, Sudan II, 4-phenylazoanisole.
a. Which compound would you expect to elute from the column first and which one last? Explain.
b. What solvent system will be used to elute these compounds from the column?

Solutions

Expert Solution

In traditional column chromatography, the compounds which have polar groups in their structures will have more affinity towards stationary phase (for example silica gel) and these compounds elute last in the column chromatography.

Compounds which are not having polar groups will not have any interactions with the stationary phase and these compounds elute first from the column.

chemist performed the reaction shown below. He then wanted to separate the mixture using column chromatography. In his protocol he used silica as the stationary phase and a mixture of hexanes/ethyl acetate (75:25). Benzene -----> (HNO3/H2SO4) --> Nitrobenzene, Dinitrobenzene Trinitrobenzene 1. In order to separate the two dyes bound the column a solution is needed with a substance that has higher affinity for the column than the dyes. Isopropyl alcohol is such a substance. If a solution of water with 70% isopropyl alcohol is applied all the purple dye elutes (comes off) the column and the dyes do not separate. A student loaded a mixture onto a small flash chromatography column in 1 mL of ethyl acetate, then proceeded to elute with hexanes/ethyl acetate 10:1. He/she found that all of the mixture came off in the first 3 fractions, with no separation. What technical mistake did the student make?

1 A) When making the decision to use column chromatography as a separation method you must consider the sample size as well as well as its chromatographic behavior (ability to be separated using an appropriate stationary and mobile phase).
