When starting with OMP and the other needed materials, describe details for the biosynthesis of dTMP
Orotate monophosphate (OMP) is formed by the addition of PRPP (phosphoribosyl pyrophosphate) to pyrimidine base called as orotic acid. This OMP is subsequently decarboxylated to UMP ( uridine monophosphate). This UMP is double phosphorylated ist by uridylate kinase and then by nucleoside diphosphate kinase to yield UTP ( uridine triphosphate). The amination of UTP results in the synthesis of CTP (cytidine triphosphate) by the action of CTP synthetase. These Uridine nucleotides are the precursors for denovo synthesis of thymine nucleotides. Thymine nucleotides are derived from denovo synthesis of dUMP (deoxyuridine monophosphate). dUMP is formed by the action of ribonucleotide reductase on UDP. dTTP synthesis requires dUMP derived from the metabolism of UDP or CDP. This dUMP is converted to dTMP by the action of thymidylate synthetase. The methyl group is donated by N5 , N10 methylene THF. thymidylate synthetase is also responsible for conversion of DHF ( dihydrofolic acid ) to THF. dTMP is subsequently phosphorylated toto dT.
