6. If you created a mutant strain of mice that were not able to increase the concentration of CstF in B-lymphocytes following antigen activities of B-cell receptors (membrane bound antibodies), how would this impact antibody production and why?

Solutions

Expert Solution

According to the given question, class swithching is a necessary process by which IgM antibody produced during primary response get change in type and quality of antibody produced in the secondary response. This class swithching process is under genetic control by CstF. All B -lymphocyte start productive life as immature cells engaged in synthesis of mIgM. Mature B- cells express both mIgM and mIgD. Both are encoded on the same mRNA, and the decision to translate IgM and IgD heavy chains is made at the level of RNA splicing.During this switching process, their is only the Ch gene segment ; the same Vh gene segment continuous to be expressed. That's was why if their is mutation in CstF or decrease in concentration , it directly affects the antibody maturation and production.

You just isolated a new strain of mutant mice, and preliminary mating suggest that the new mutant phenotype is not inherited in an autosomal fashion. Circumstantial evidence seems to indicate that the mutant phenotype may be following either an X-linked dominant or a mitochondrial-type inheritance pattern. a.) What two informative crosses would you set up to distinguish between these two possibilities? b.) How would the results distinguish between the two possibilities?

A given strain of mice were injected with the same amount of tumor pulp that came from a large tumor excised from another mouse. After the tumor injections, the mice were then randomly injected with one of two chemotherapy treatments (C or B). Fifty days after the chemotherapy injection, the tumor volumes (in cubic centimeters) were measured. Researchers hypothesize that treatment C will be more effective than treatment B. What can be concluded with α = 0.01? The results are...

You mate a male from a true-breeding strain of dwarf mice to a female from a true-breeding normal-sized strain. The F1 mice are all normal in size. What would the phenotypes of the F1 and F2 mice be in the reciprocal of the above cross if the dwarf character is caused by a mitochondrial genome defect?

You engineer a strain of S. cerevisiae cells that express mutant histone octomer complexes that exhibit a negatively-charged DNA-binding surface. Which of the following would you expect to happen in a sample of these cells? The chromosomes of the mutant strain would condense only during mitosis. Nucleosome core particles would not form. Epigenetic inheritance would be observed. All chromatin in the mutant strain would be condensed. The DNA would package too tightly for any gene expression to occur.

You discover a mutant yeast strain that cannot grow when provided with pyruvate as its sole carbohydrate, although it grows normally when given glucose as a carbon energy source; you call this the Pyr– trait. Wild-type yeast can grow well using either carbon source; you designate this the Pyr+ trait. You suspect that the mutant yeast have defective mitochondria. A. Why is mitochondrial function critical for growing on pyruvate? B. You want to determine whether the mutation responsible for the...

Suppose you have two strains of mice, S1 and S2. Strain S2 is genetically modified to metabolize a pharmacon P supposedly faster than S1. You conducted an experiment in a sample set of each strain, in which the pharmacon was injected and its concentration in blood was measured every 15min for 2h. Of course, age, gender, and weight was recorded for each animal. You want to statistically demonstrate that the metabolic rate of N is higher in S2 than S1....

Wild-type E. coli can grow on minimal medium. You have isolated a mutant strain of E. coli that grows poorly in minimal medium that contains lactose and arabinose and lacks glucose. In ONPG-containing minimal medium lacking glucose the mutant strain produces very low levels of b-galactosidase activity in both the absence and presence of IPTG. The strain does not grow well when transformed with a plasmid containing the lacZ and lacY genes. The strain also does not grow well when...

Predict what will happen to the expression of β-galactosidase (lacZ gene) in a mutant strain lacking lacI-gene (cells are not able to produce LacI-protein): A. lacZ gene will not be expressed. B. lacZ gene will be expressed only when both lactose and glucose are present. C. lacZ gene will be expressed only in the presence of lactose; maximum expression will be observed in the absence of glucose. D.lacZ gene will not be repressed; maximum expression will be observed in the...
