discuss chromatography as a method of chemical analysis
First, we need to understand what is Chemical analysis?
Chemical Analysis is a series of process to identify or to quantify a chemical substance from a mixture and to identify the structure of each chemical compounds.
Chromatography is a chemical analysis through which we can separate a mixture.
Common chromatography used in laboratory / industrial lab are:
1. Thin layer chromatography (TLC) (used to separate non-volatile mixtures of chemical compound)
2. Column chromatography (used to separate chemical compound individually from a mixture of chemical compound)
In column chromatography, we have to pack the column with a mixture of silica gel (mesh size: 60-120/100-200/200-300) and solvent (hexane or DCM). We will make the slurry of the compound (which need to be separated) and poured in the column. Further, we will pour percentage solution (Hexane: Ethyl acetate) to it. Depend upon the polarity of compound, the mixture will be separated through column chromatography.
