Q. What is CactSu? How was it discovered and what is the molecular change (in general terms) in CactSu comparing to the normal version of maternal Cactus?

  • CactSu is a gain of function mutant allele of cactus
  • Cactus is involved in the inhibition of the nuclear translocation of Dorsal protein by binding Dorsal and retaining in the the cytoplasm.
  • CactSu allele is a strong dominant suppressor of ventrilization but it doesnot have any phenotype of its own and flies are fully viable with females producing wild type embryo. CactSu encodes a slightly truncated protein as compared to wild type Cactus protein and also appears more stable.
  • CactSu/CactSu embryos 20% of the nuclei in the embryo circumferance had high levels of Dorsal. This percentage is reduced as compared to 27% in th wild-type embryos.
  • It was discovered as a gain of function mutation (that were no longer able to suppress ea 831 and mapped to the same site as Su) during a genetic screen to revert the suppressing effect of the mutation.
