The nicotinic receptor binds to acetylcholine and is cholinergic ionotropic receptor. This receptor does not use secondary messengers. Instead the receptor is linked to ion channels. Stimulation of the receptors will cause influx of ions into the cell due to opening of the ion channels. Sodium, potassium and sometime even calcium influx is seen, which then activates downstream signaling cascades such as PI3K signaling.

The other receptors-alpha 1 and 2, beta 1 and 2 are all G protein coupled receptors. Binding of the ligands to these receptors, triggers the hydrolysis of G protein to form G alpha subunit, which then activates adenylyl cyclase to form cAMP. Thus, they are all use secondary messengers for activity.
