Both spread plate and pour plate methods used to isolate pure colony , depending on the using type bacterial culture and amount of inocum and purpose of study with bacteria method is choosen

generally pour plate is used to isolate and identify aerobes, anaerobes,or facultative aerobes,and also allow the growth of microaerophills, because in this method bacteria grows in side the media and also on the surface of medium.

pour plate method the plate is prepared by molten agar is poured on the(1 ml ) inoculum in the Petri dish and gently swirrled ,here bacteria get more area to grow, allows easy identification of bacteria.

spread plate method is used to count and isolate bacterial colonies

on the surface of the agar, inocum spread on the surface of solidified agar on plate with spreader, here only 0.1 ml of inocum is added ,less area to grow when compared to pour plate method but perform even distribution of colonies and isolate specific clonal colonies
